Broad-spectrum Antiviral siRNAs: Influenza Strains Targeted by Anticipation

Project description

Broad spectrum therapies against influenza

Fighting emerging and constantly evolving viruses, such as influenza, presents significant challenges, especially given the inefficiency of current influenza vaccines, which often fail to provide protection against new strains. The ERC-funded BARISTA project aims to develop a novel antiviral therapeutic that remains effective across a wide range of influenza strains, including future ones. The approach will involve small interfering RNAs (siRNAs) to silence genes expressed in a variety of influenza strains. These siRNAs will be structurally optimised and tested for their broad-spectrum activity in various influenza models. The goal is to provide a therapeutic approach to combat emerging viruses and improve responses to future outbreaks.

Objective

Fighting emerging viruses has been a major challenge in the development of novel therapeutics. With the current global changes and massive modification of the biosphere, the issue will surely increase in the future. To overcome this challenge, we will demonstrate on influenza that we are capable to produce active antiviral against a strain that is not yet existing. The BARISTA project will allow to develop a novel therapeutic approach allowing to preserve therapeutic activities over a wide range of influenza strains and we will demonstrate its efficacy on the influenza strain of the next outreach. This achievement will be obtained by guiding the design of chemically optimized siRNA by detailed NMR structural analysis. Those molecules will be tested both in vitro and ex vivo and their broad-spectrum activity will be demonstrated on past, present and future influenza strains. This project will therefore open the way to therapies that can be designed and optimized, even before the appearance of a novel strain, providing an invaluable help in fighting emerging viruses.

Programme(s)

Multi-annual funding programmes that define the EU’s priorities for research and innovation.

Topic(s)

Calls for proposals are divided into topics. A topic defines a specific subject or area for which applicants can submit proposals. The description of a topic comprises its specific scope and the expected impact of the funded project.

Funding Scheme

Funding scheme (or “Type of Action”) inside a programme with common features. It specifies: the scope of what is funded; the reimbursement rate; specific evaluation criteria to qualify for funding; and the use of simplified forms of costs like lump sums.

Net EU contribution

Net EU financial contribution. The sum of money that the participant receives, deducted by the EU contribution to its linked third party. It considers the distribution of the EU financial contribution between direct beneficiaries of the project and other types of participants, like third-party participants.
