Host microbe dialogue through metabolic interactions

Objective

The intestinal microbiome is a complex community of organisms that exists in symbiosis with its host. Microbiota perform essential digestive, metabolic, and immunomodulatory functions in health, and disturbance of the microbiota can contribute to disease. Microbial metabolites are a key ingredient in healthy host-microbe dialogue, bolstering barrier immunity and immune regulatory pathways. Our recent work on microbiota-derived small molecule metabolites ADP-heptose and the short chain fatty acid, butyrate, has identified new immune regulatory and microbicidal functions that could be targeted in inflammatory bowel disease. However, harnessing metabolites as disease therapeutics has been challenging because of their diverse cellular targets and the complex bacterial community ecology governing their production and function. Using our recent technological advances we propose a multi-disciplinary approach spanning the frontiers of immunology, cell biology, gnotobiology, microbial ecology and computational biology, to deconvolute host microbiota interactions over space and time. Using ADP-heptose and butyrate as exemplars, we will use metabolite-centric designer microbiome communities together with integrated spatial, omics-scale data in tissue sections, to chart the passage of microbiota-derived metabolites into tissue niches to reveal their cellular targets and influence on transcriptomic and proteomic networks relevant to immune regulatory and microbicidal function. In addition to our in vitro-defined cellular pathways, this will identify novel interkingdom targets and opportunities to treat disease niches, leaving other metabolic pathways and cellular interactions unaffected. Results of these studies will be used to design new vaccine enhancing strategies and more specific therapeutic approaches in IBD patients.

Programme(s)

Multi-annual funding programmes that define the EU’s priorities for research and innovation.

Topic(s)

Calls for proposals are divided into topics. A topic defines a specific subject or area for which applicants can submit proposals. The description of a topic comprises its specific scope and the expected impact of the funded project.

Funding Scheme

Funding scheme (or “Type of Action”) inside a programme with common features. It specifies: the scope of what is funded; the reimbursement rate; specific evaluation criteria to qualify for funding; and the use of simplified forms of costs like lump sums.

HORIZON-ERC - HORIZON ERC Grants

Call for proposal

Procedure for inviting applicants to submit project proposals, with the aim of receiving EU funding.

THE CHANCELLOR, MASTERS AND SCHOLARS OF THE UNIVERSITY OF OXFORD
Net EU contribution

Net EU financial contribution. The sum of money that the participant receives, deducted by the EU contribution to its linked third party. It considers the distribution of the EU financial contribution between direct beneficiaries of the project and other types of participants, like third-party participants.
