Objective
The heart’s mechanical workload drives cardiomyocyte biology - shaping proliferation, sarcomere organization, and function - yet our ability to modulate it with precision has been limited. CARDIOSWITCH will change this by integrating stem cell–derived models, human cardiac slices, and preclinical transplantation with novel chemo- and optogenetic tools, plus targeted sarcomere disassembly, to reversibly control cardiomyocyte physiology in space and time. Aim 1 will determine how continuous mechanical workload constrains cardiomyocyte proliferation, using precise, reversible modulation of contractility to isolate cause-and-effect. Aim 2 will define the quantitative relationship between the number of actively contracting cardiomyocytes and overall heart function - a critical but unexplored parameter for all regenerative strategies. Aim 3 will apply these tools in preclinical congenital heart disease models, testing stem cell-derived cardiomyocyte transplantation and strategies to overcome its major translational hurdle: ventricular arrhythmias. With expertise in cardiac tissue engineering, opto- and chemogenetics, and preclinical congenital heart defect models, I am uniquely positioned to deliver these advances. If successful, CARDIOSWITCH will deepen our understanding of heart biology and provide a foundation for regenerative therapies that restore synchronised function in children with severe congenital heart defects.

Funding Scheme
Funding scheme (or “Type of Action”) inside a programme with common features. It specifies: the scope of what is funded; the reimbursement rate; specific evaluation criteria to qualify for funding; and the use of simplified forms of costs like lump sums.

Net EU financial contribution. The sum of money that the participant receives, deducted by the EU contribution to its linked third party. It considers the distribution of the EU financial contribution between direct beneficiaries of the project and other types of participants, like third-party participants.
