A Pan-population study of obesity, risk and metabolic architecture

Objective

What am I going to do?
I will use data from three large prospective studies in East Asia, Europe and Latin America to relate adiposity measures with >1000 human diseases, assess cause-effect associations, clarify underlying mechanisms, discover molecular signatures of adiposity, explore ancestry differences, and identify potential drug targets for treatment of obesity and its associated diseases.
Why am I going to do this?
Obesity is one of the most important global public health challenges of this century. Despite substantial previous research on the causes and consequences of obesity, the true disease burden attributed to adiposity and the underlying mechanisms, and relative importance of adiposity subtypes are not fully understood. Moreover, the extent to which these differ by ancestry is not yet properly established. Likewise, little is known about the reasons why many lean or normal weight people develop metabolic diseases, and elucidation of molecular signatures that can help identify lean individuals who are molecularly obese (or vice versa) will be informative. Using an integrated approach, this project will provide important new evidence to evaluate the epidemiology of obesity and develop precision health approaches for disease prevention in diverse populations.
How am I going to do this?
This proposal will conduct detailed analyses of data from three prospective studies involving >1.15 million individuals from three continents, with proper assessment of adiposity, well-characterised disease outcomes, cohort-wide genetic data, and deep molecular profiling. Both conventional and novel advanced analytical methods will be applied in innovative experimental paradigms across four distinct work packages that utilise high-dimensional phenotype, genetic and omics data, with due consideration of data standardisation and harmonisation, study power, bias, multi-testing, causal inference, and cross-validation to ensure robust, generalisable and impactful findings.
