Objective
Mucosal barriers are constantly exposed to fungi that must be killed and cleared to prevent severe infection and chronic illness. Pathogenic fungi germinate into large filamentous hyphae that induce the deployment of neutrophil extracellular traps (NETs), leaving behind a substantial debris of dead hyphae and cytotoxic NETs. These components are highly proinflammatory and must be cleared to resolve inflammation. The mechanisms that digest dead hyphae and their contribution to disease are unknown. We discovered a novel neutrophil effector mechanism that deposits immunomodulatory factors on dead hyphae. We propose that this novel cell biological phenomenon we named “neutrophil painting”, enables hyphal viability sensing to regulate inflammation and hyphal clearance. Moreover, we uncovered the first known mechanism that digests dead hyphae in macrophages. We will characterise the molecular mechanism of neutrophil painting and the processes that digest hyphae in macrophages. We will investigate how neutrophil painting controls hyphal digestion in macrophages and explore how these pathways regulate inflammation and fungal debris clearance during infection in vivo. This work will reveal a novel mode of immune communication that utilizes pathogen-bound signals, unveil novel facets of phagocyte cell biology, and highlight the importance of dead fungal clearance for health.

Funding Scheme
Funding scheme (or “Type of Action”) inside a programme with common features. It specifies: the scope of what is funded; the reimbursement rate; specific evaluation criteria to qualify for funding; and the use of simplified forms of costs like lump sums.

Net EU financial contribution. The sum of money that the participant receives, deducted by the EU contribution to its linked third party. It considers the distribution of the EU financial contribution between direct beneficiaries of the project and other types of participants, like third-party participants.
