Objective
Multicellular organisms host diverse microbes that play vital roles in their health and crop resilience. The host and its microbes form intricate ecosystems characterized by heterogeneity and multidimensionality, hindering the mechanistic understanding needed for targeted management. The ambitious challenge is to disentangle the complexity of signalling events involved in plant root-microbiota assembly to achieve an optimal cost-benefit outcome from these cross-kingdom alliances. The central hypothesis is that a crosstalk between different microbe-induced signalling pathways is established and governs the formation of environmental micro-niches on or inside the root where specific microbial infections occur. ProMIC will identify plant genes and pathways that coordinate the formation of these niches in space and time. We will pioneer innovative approaches based on synthetic gene circuits, activation of targeted pathways, and tailored microbial infections. These will be combined with state-of-the-art bacterial genetics, spatial omics, and advanced cell biology to study the intricate signalling networks driving microbiota assembly at an unprecedented level of granularity. Lotus japonicus is our testbed, as it engages in multiple symbioses and associations with diverse soil microbes. ProMIC capitalizes on the substantial resources for genetic and molecular studies of host-associated microbiota in Lotus to establish a framework for understanding how multicellular organisms host complex microbial communities, a stepping stone for the efficient use of microbial solutions in sustainable crop production. ProMIC builds on my well-established expertise in plant-microbe interactions of varying complexity and on our fundamental discoveries from receptor signalling studies to provide molecular knowledge that complements the current systems-level exploration of the field.
