GENOME ENGINEERING OF MACROPHAGES TO MEDIATE REGENERATION OF THE FIBROTIC LUNG

Objective

Idiopathic Pulmonary Fibrosis (IPF), affects over 100,000 people in Europe and more than 3 million globally, causing scarring of lung tissue, respiratory decline and death. IPF remains a serious unmet medical need with limited treatment options. Genetic engineering of macrophages to create an innovative sense and respond-based cellular therapy offers a new solution. BREATH will develop functionally improved macrophages, genetically designed and enhanced to counteract dysfunctional macrophages in the fibrotic lung, restoring immune balance and halting fibrosis effectively. BREATH will harness modern synthetic biology concepts and integrate state-of-the art knowledge in genetic switches to generate genetically modified human macrophages using known IPF gene targets and innovative strategies for tight control of therapeutic genes. Simultaneously, we will accelerate the discovery and implementation of new pro-regenerative synthetic gene circuits for the treatment of IPF through gene silencing. The cell products will be evaluated for efficacy and safety using state of the art in vitro and ex vivo models of the human lung and in vivo, using pre-clinical mouse models of fibrosis. To facilitate the clinical translation of the products developed, we will carry out a feasibility pilot study in healthy volunteers, together with a programme of clinician, patient and regulatory engagement. Furthermore, we will develop a stratification framework and collect and integrate the information required to inform future clinical translation. By positioning genetically enhanced macrophages as a new class of regenerative medicine, BREATH will, in the long term, reduce the societal and economic burden of IPF and pave the way for other innovative therapies for fibrotic diseases including the liver and heart.

Fields of science (EuroSciVoc)

CORDIS classifies projects with EuroSciVoc, a multilingual taxonomy of fields of science, through a semi-automatic process based on NLP techniques. See: The European Science Vocabulary.

Keywords

Project’s keywords as indicated by the project coordinator. Not to be confused with the EuroSciVoc taxonomy (Fields of science)

Programme(s)

Multi-annual funding programmes that define the EU’s priorities for research and innovation.

Topic(s)

Calls for proposals are divided into topics. A topic defines a specific subject or area for which applicants can submit proposals. The description of a topic comprises its specific scope and the expected impact of the funded project.

Funding Scheme

Funding scheme (or “Type of Action”) inside a programme with common features. It specifies: the scope of what is funded; the reimbursement rate; specific evaluation criteria to qualify for funding; and the use of simplified forms of costs like lump sums.

Call for proposal

Procedure for inviting applicants to submit project proposals, with the aim of receiving EU funding.

Net EU contribution

Net EU financial contribution. The sum of money that the participant receives, deducted by the EU contribution to its linked third party. It considers the distribution of the EU financial contribution between direct beneficiaries of the project and other types of participants, like third-party participants.
