Objective
IMPACT will revolutionize the manufacturing of Chimeric Antigen Receptor (CAR) Cell therapies, accelerating access to life-saving treatments for blood cancers and immune disorders. While CAR therapies have shown high efficacy in treatment-resistant haematological malignancies, their adoption is limited by costly, slow, and manual manufacturing processes, inconsistent batch quality, and constrained lentiviral vector (LV) supply. IMPACT addresses these barriers through a digitally integrated, scalable, and patient-adaptable CAR Cell production platform.
A 15-partner consortium across 9 countries will co-develop innovations to transform every stage of the value chain. LV production will be scaled tenfold using closed, fixed-bed bioreactors with sustainable purification and stabilization. Quality control will be accelerated by nanotechnology-enabled single-particle sensing, enabling real-time, label-free potency assessment. Patient-specific efficacy and safety will be evaluated using an integrated suite of Organ-on-Chip models—Cancer-on-Chip, Bone Marrow-on-Chip, and Blood-Brain Barrier—reducing reliance on animal testing. These models will enable predictive readouts of therapeutic performance and side effects, ensuring safer, faster, and more personalized treatment decisions. IMPACT also fosters skills development and workforce training through cross-sector collaboration, ensuring the technologies are adopted effectively across Europe.
Central to the project is a real-time, AI-driven digital twin of the entire manufacturing process, integrating bioprocess data, QC, and in vitro readouts to enable adaptive control and traceability in line with GxP standards. Green solvents, reduced plastic use, and full life-cycle assessments embed sustainability throughout.
IMPACT will deliver a robust, interoperable platform that shortens production time, lowers cost, and broadens patient access, establishing a European blueprint for next-generation ATMP manufacturing.
