Objective
Alzheimer’s disease (AD) is a progressive neurodegenerative disorder and the most common cause of dementia, characterized by memory loss, cognitive decline, and behavioral changes. It affects over 55 million people worldwide, with nearly 10 million new cases diagnosed each year. As global populations age, prevalence is expected to rise sharply, placing immense pressure on healthcare systems and economies. AD is a major public health challenge with no cure, and no effective treatment to stop disease progression.
Decades of research on AD have centered on amyloid-β (Aβ), yet this has proven insufficient to explain AD multifaceted cytopathology. One common denominator in the onset of AD pathology is synaptic dysfunction and network hyperexcitability. My lab recently uncovered that APP-CTFβ, the immediate precursor to Aβ, localizes at synapses where it appears to regulate excitatory function. Strikingly, over-accumulation of APP-CTFβ triggers synaptic dysfunction and network hyperexcitability. Importantly, we found elevated levels of APP-CTFβ in post-mortem brain of AD patients. Could this be the missing piece of the puzzle?
This project will develop new molecules names “OligoBlockers” targeting APP-CTFβ instead of Aβ as a novel avenue to prevent AD at an early stage.

Net EU financial contribution. The sum of money that the participant receives, deducted by the EU contribution to its linked third party. It considers the distribution of the EU financial contribution between direct beneficiaries of the project and other types of participants, like third-party participants.
