Objective
Infection burden in childhood is a leading global cause of morbidity and mortality, but it remains a clinical challenge to identify why otherwise healthy children are more prone to infections than others and how to prevent these as this is an unmet medical and societal need. The aim of IDOL is to investigate infectious diseases in childhood with the ultimate goal of developing personalized preventive strategies by understanding host and environmental risk factors, underlying mechanisms and detect biomarkers using longitudinal proteomic and metabolomic data layers in multi-omics analyses and testing these in experimental models using human bronchial cells. This project is a collaboration between leading experts within the field based on 5 birth cohorts across the world in Denmark (University of Copenhagen), Sweden (Karolinska Institute), Canada (University of Toronto) and United States (Harvard Medical School). The interdisciplinary group lead by the applicant consisting of professors in pediatrics, statistics and environmental health as well as wet lab collaborators for the mechanistic studies will bridge basic and clinical sciences. I will utilize data from more than 2200 children from birth cohorts having an extensive amount of information on early environmental risk factors including detailed information on daily infection symptom load, medications, several omics data layers including blood metabolomics, proteomics and immunology measures data as well as genotyping of the children. I include the only two birth cohorts having unique data on infant lung function measurement before the first infection episode. Further, I utilize data from Danish national registries to perform sibling design studies investigating the influence of prenatal antibiotic exposure on offspring infection risk. Hence, this project is uniquely positioned to detect novel infectious disease biomarkers and investigate early life risk factor mechanisms towards a personalized prevention.
