The Gastrointestinal Biocorona: The Missing Link in Lipid Digestion and Oral Drug Delivery

Objective

Every foreign material that enters the human body is rapidly covered by biomolecules. This ‘biocorona’ has revolutionized our understanding of drug delivery in plasma. Yet, it has rarely been studied for one of the most complex biochemical environments: the gastrointestinal (GI) tract. Evidence for GI-corona formation on lipids and lipid-based drug delivery systems (LBDDSs) is still missing to date, although it could be equally transformative for our understanding of lipid digestion and oral drug delivery.

GI-CORONA will i) provide the first quantitative composition of the GI-corona on dietary lipids and LBDDSs, ii) unravel its hidden effects on lipid digestion, and iii) optimize LBDDS formulations by accounting for the GI-corona. The recent advances in proteomics will be exploited to gain quantitative insights in GI-corona composition from the gastric to the intestinal stage. Current lipid digestion and drug delivery models, which omit most GI biomolecules, will be refined with relevant GI biomolecules to provide more predictive digestion models. Ultimately, the drug delivery potential from LBDDSs will be optimized accounting for GI-corona formation using biorelevant in vitro intestinal multi-cell models.

The expected outcomes are: (i) a robust methodology for corona analysis on liquid lipids, ii) the first comprehensive dataset of GI-corona protein and lipid composition; (iii) next-generation in vitro digestion models with higher predictive power; and (iv) optimized design criteria for LBDDSs for more effective oral drug delivery. By uncovering this hidden layer of biological interaction, GI-CORONA will transform our understanding of lipid digestion and oral drug delivery, bridging fundamental biophysics, nutrition, and pharmaceutical science.

Fields of science (EuroSciVoc)

CORDIS classifies projects with EuroSciVoc, a multilingual taxonomy of fields of science, through a semi-automatic process based on NLP techniques. See: The European Science Vocabulary.

Keywords

Project’s keywords as indicated by the project coordinator. Not to be confused with the EuroSciVoc taxonomy (Fields of science)

Programme(s)

Multi-annual funding programmes that define the EU’s priorities for research and innovation.

Topic(s)

Calls for proposals are divided into topics. A topic defines a specific subject or area for which applicants can submit proposals. The description of a topic comprises its specific scope and the expected impact of the funded project.

Funding Scheme

Funding scheme (or “Type of Action”) inside a programme with common features. It specifies: the scope of what is funded; the reimbursement rate; specific evaluation criteria to qualify for funding; and the use of simplified forms of costs like lump sums.

HORIZON-ERC - HORIZON ERC Grants

Call for proposal

Procedure for inviting applicants to submit project proposals, with the aim of receiving EU funding.

UNIVERSITE DE FRIBOURG
Net EU contribution

Net EU financial contribution. The sum of money that the participant receives, deducted by the EU contribution to its linked third party. It considers the distribution of the EU financial contribution between direct beneficiaries of the project and other types of participants, like third-party participants.
