INNOVATIVE RATIONAL PROMOTER BREEDING FOR CLIMATE RESILIENT CROPS INPROBED

Objective

INPROBED pioneers a paradigm shift in plant biotechnology by moving from conventional gene editing to precision promoter engineering, by adapting natural gene regulation to build stress resilience and nutritional quality into food crops. The project will develop the first predictive “promoter design platform” for tomato, integrating single-cell MOA-seq with deep-learning models to map and reprogram cis-regulatory elements that control plant responses to heat, drought, and micronutrient limitation. This approach enables precise, predictable, and regulation-compatible genome edits that enhance resilience without compromising growth or yield.

Using CRISPR-based promoter editing, INPROBED will generate tomato lines that accumulate more zinc, iron, and provitamin D₃ while maintaining stable performance under stress. These lines will be validated through multi-omics analysis and high-throughput phenotyping, resulting in pre-breeding of proof-of-concept lines for semi-field trials.

The consortium unites world-leading expertise in genomics, cistromics, and translational crop science: Heinrich Heine University advances promoter discovery and single-cell analysis; Wageningen University and the Weizmann Institute lead nutritional and metabolic engineering; KeyGene leads the genome editing, single cell technology development and ensures rapid integration into breeding pipelines; and PLANeT Biotech drives phenotyping, validation, and commercialization.

INPROBED is a generic technology platform enabling crop improvement via rational promoter engineering in a crop-agnostic manner.
