Next-generation therapeutics for vascular diseases

Objective

TIE2gether is a multidisciplinary research and training network addressing major unmet needs in vascular medicine.

A wide range of vascular-related diseases—including chronic kidney disease, diabetic vasculopathy, sepsis, glaucoma and vascular malformations—affects millions and imposes a major societal and economic burden. Yet these conditions still lack effective, mechanism-based therapies. A unifying feature across these clinically distinct diseases is dysregulation of the TIE2 signalling cascade: hyperactivation drives vascular malformations while insufficient activity causes vascular leak, inflammation and organ dysfunction. Restoring balanced TIE2 signalling thus represents a major, largely unexplored therapeutic opportunity with relevance across both common and rare diseases.

TIE2gether translates this opportunity into real-world benefit by training a new generation of scientists capable of bridging fundamental discovery with therapeutic innovation. The network brings together leading experts in vascular biology, organ-on-chip technologies, advanced disease modelling, chemical biology, protein engineering, and drug discovery. By centring around a single, highly druggable pathway, TIE2gether creates strong synergy and accelerates the transfer of mechanistic insight into validated therapeutic strategies evaluated in human-relevant systems.

Fourteen Doctoral Candidates will gain cutting-edge competences in dissecting TIE2 signalling mechanisms, developing state-of-the-art disease models and designing and assessing next-generation therapeutics, including network-developed drug leads. Complemented by interdisciplinary, clinical and patient-facing training, they will acquire the innovation mindset, translational expertise and transferable skills needed across academia, industry and the biomedical sector.
