Objective
LOCAL-CAR is a Proof-of-Concept project aimed at validating a modular hydrogel-based platform for localized mRNA immunotherapy, targeting peritoneal tumor burden in ovarian and colorectal cancer models. Building on ERC-funded technologies – ABGel and aNP-mRNA – the project integrates these into a single ABGel-mRNA system for sustained intraperitoneal immunotherapy. The platform will generate HER2-CAR-expressing macrophages (CAR-M) directly within the peritoneal cavity, enabling local anti-HER2-tumor activity.
The project objectives are: (1) optimize ABGel-mRNA formulations encoding HER2-CAR constructs and screen their ability to generate functional CAR-M in vitro; (2) evaluate in vivo CAR‑M induction and therapeutic efficacy in mouse ovarian and colorectal cancer models; (3) develop IP strategy and expand patent portfolio; (4) conduct market analysis and engage stakeholders in the emerging in vivo CAR therapy landscape; and (5) disseminate findings and build strategic partnerships. Co-applicant BioTrip supports IP management, regulatory strategy, and commercialization. LOCAL-CAR addresses a critical unmet need in oncology and offers a scalable, biocompatible solution with broad translational potential in immunotherapy, autoimmune diseases, and vaccination.

Net EU financial contribution. The sum of money that the participant receives, deducted by the EU contribution to its linked third party. It considers the distribution of the EU financial contribution between direct beneficiaries of the project and other types of participants, like third-party participants.
