Bioactive Peptide-based Hydrogels for Alveolar Bone Regeneration

Objective

Tooth loss and the resulting alveolar bone resorption are highly prevalent conditions, particularly in aging populations and in patients suffering from inflammatory periodontal diseases. Dental implants are the preferred solution for replacing missing teeth; however, successful implantation requires sufficient alveolar bone volume. Bone resorption frequently leads to unfavorable conditions of the alveolar ridge, making socket preservation and bone augmentation procedures necessary to regenerate sufficient bone volume prior to dental implant placement. Current bone grafting approaches remain limited, particularly in elderly patients, diabetics, and smokers, highlighting the urgent need for improved regenerative strategies.
Here, we aim to develop a novel alveolar bone graft based on our patent-protected polysaccharide-peptide composite hydrogel, developed within the PersonalBone ERC-StG project. Unlike conventional bone graft materials, which primarily function as osteoconductive scaffolds, our technology is designed as an osteoinductive, immune-instructive biomaterial that actively recruits and modulates the host immune response to promote bone regeneration. By creating a bioactive microenvironment that stimulates immunomodulation and osteogenesis, this material represents a paradigm shift in alveolar bone regeneration strategies.
Within this ERC-PoC project, we will optimize the hydrogel formulation to achieve the mechanical stability required for socket preservation and its integration with clinically used bone graft particulates. The optimized composite graft will be evaluated for biocompatibility, bone regeneration capacity, and resorption kinetics in critical-size bone defect models in vivo, with clinical input from leading dentists. This technology has the potential to significantly advance current bone augmentation treatments and improve clinical outcomes for patients requiring dental implants.
