Innovative exploitation of PVAT-vascular crosstalk through integrated tissue biology, animal models, and organ-on-chip to deliver clinically validated ASCVD biomarkers

Objective

Cardiovascular disease (CVD) remains the leading cause of mortality in Europe, driven by unresolved vascular inflammation and the lack of mechanistically grounded biomarkers. INNOVATOC addresses this challenge by investigating perivascular adipose tissue (PVAT) as a key regulator of vascular inflammation and plaque instability through a structured programme of international, intersectoral and interdisciplinary knowledge exchange.
The project will generate a unique cross-species PVAT–plaque multi-omics dataset from human tissues and complementary animal models. Integrated analysis, supported by single-nuclei RNA sequencing, will enable identification and prioritization of mediators involved in PVAT–vascular crosstalk and inflammation resolution, followed by functional validation in advanced adipocyte–vascular–immune interaction systems.
INNOVATOC will deliver: 1) prioritised panel of 15 candidate biomarkers (miRNA, protein and specialised pro-resolving lipid mediators, SPLM), 2) validated LC–MS/MS-based assay for targeted SPLM quantification, designed for scalability and clinical applicability, 3) novel organ-on-chip (OoC) prototype that recapitulates PVAT–vascular interactions under physiologically controlled conditions. These outputs will support the establishment of a predictive biomarker panel for cardiovascular risk prediction, validated in well-characterised clinical cohorts.
Through structured secondments and joint activities, INNOVATOC will enable intensive knowledge transfer across disciplines and sectors, fostering innovation in biomarker discovery, analytical technologies, and organ-on-chip systems. The consortium brings together 6 academic institutions, 5 non-academic partners (including 2 SMEs and 3 clinical centres), and 1 international associated partner, ensuring balanced expertise across research, clinical translation, and technological development, and supporting sustainable long-term collaboration networks.
