Drug therapy is central to the treatment of most diseases. The response to drugs varies widely between individuals. Part of this variability is due to genetic differences between individuals. The TransporterPGx-project aimed to systematically search for functionally significant genetic variants in drug transporters. Drug transporters are membrane proteins which mediate the influx of drugs into cells and efflux of drugs out of cells, and can therefore impact the concentrations of drugs in tissues important for their beneficial effects, but also for adverse effects. Within the project, several novel genetic variations were discovered in drug transporters and also in drug metabolizing enzymes. These variations may be useful in the future for personalizing drug therapies.
