Objective
Chronic inflammatory disease affects almost one third of the EU population and an increasing proportion of this burden is made up of liver disease. Large increases in death rates from chronic liver disease and cirrhosis have occurred in many EU countries and are predicted to continue. Chronic inflammation and cirrhosis develop in response to liver injury mediated by a wide range of reagents from hepatitis viruses, to alcohol and autoimmune processes.
In all cases, persistent inflammation drives the progression and severity of disease but there are currently no effective therapies that modulate or switch off chronic inflammation. There is therefore a pressing need for a greater understanding of the mechanisms of chronic inflammatory liver disease as well as for the development of new therapies. The need is best addressed through action at a Community level to establish and maintain long term collaborative studies aimed, in the long term, at the development of new drugs.
The applicants have demonstrated a critical role for the endothelial adhesion molecule VAP-1 in the recruitment of effector leukocytes to the liver and that it is the recruitment of effector leukocytes that sustains chronic liver inflammation. One of the applicants has developed novel therapeutic reagents based on antibodies and small molecule drugs to inhibit VAP-1 function.
The current proposal has the objective to consolidate and further develop a partnership between the groups using the expertise in the academic group to explore new avenues in VAP-1 research and test candidate anti-VAP-1 therapies developed by the SME in both human in vitro and animal in vivo studies. This will be facilitated by the exchange of knowledge between the partners. The partnership will also strengthen the European biomedical science base by improving the competitiveness of a European SME.
