Applications of NMR spectroscopy to the study of the interactions of glyconanoparticles with other biomolecules as models of multivalent biological molecular recognition processes

Objetivo

The project presented here aim for the development and application of NMR spectroscopy techniques for the characterization of biologically relevant molecular recognition events mediated by carbohydrates using glyconanoparticles (gold nanoparticles functionalised with synthetic oligosaccharides) as multivalent systems in carbohydrate-carbohydrate and carbohydrate-protein interactions. These glyconanoparticles (GNPs) have been shown to be very useful for the study of molecular recognition processes, yet their accessibility by NMR spectroscopy has remained elusive, due to their particular spectral properties. Here it is proposed the use of advanced NMR techniques (HR-MAS, DOSY) for their characterization as free particles in solution, and to apply classical and recently developed NMR experiments (HR-MAS STD NMR, Water-LOGSY, Diffusion NMR, trNOE) to investigate their interactions with other relevant biomolecules.
