Participation of endomembranes and components of the cell wall on the maintenance of plant cell polarity

Objetivo

The mechanisms of tip-growth have been studied thoroughly for a long time, but the complex view is still far from complete. The main questions asked by scientists from different fields of cell biology are how tip growth is regulated, what is the principle of proper "anchoring" of protein complexes required for root hair formation and what signalling background is involved in root hair tip growth. The aim of our project is the characterization of the integrity of growing tips by modern cell biological method s like live cell imaging, phenotyping of transgenic plants and mutant screening.

This complex approach will be focused to determination of cell wall - plasma membrane interactions, sterol distribution in the plasma membrane, behaviour of structural components in the cell wall and the endocytosis in different developmental stages of root hair formation. Special attention will be paid to basic developmental checkpoints of root hairs under influence of changing environmental stress conditions. Experiments will be made in a multidisciplinary approach in collaboration with two other European Institutions by a combination of several cell biological and genetic techniques. Results will contribute to the understanding of regulated cooperation between cell wall, plasma membrane and intracellular signalling components determining root hair tip growth and plant cell polarity.
