Objective
Cytokines and their receptors play a crucial role in blood formation, inflammatory responses and immune defences. Defects in cytokine receptors or in the associated janus kinases (JAKs) have been linked to chronic disease, such as severe combined immuno deficiencies, congenital neutropenia, myeloproliferative disorders, leukaemia and autoimmune diseases. Members of this RTN have been involved in the discovery of JAKs, cloning of genes encoding cytokine and cytokine receptors and identification of mutant receptors and JAKs in human diseases.
The overall objective of this RTN is to decipher the mechanisms by which JAKs affect the traffic and consequently the signalling outcomes of a number of cytokine receptors relevant to normal hematopoietic and immune functions and associated diseases. Findings revolving around the cytokine receptor-JAK scaffold will be exploited to ultimately modulate receptor levels, spatial configuration, ligand sensitivity and signalling duration towards therapeutic purposes.
To this end, we will:
(1) Share and combine expertise, knowledge, reagents and technologies to dissect common and differential mechanisms and functional consequences of cytokine receptor traffic in normal and pathophysiological conditions of the hematopoietic and immune systems,
(2) Train early stage researchers (ESR) and experienced researchers (ER) in state-of-the art cytokine receptor biology and multifaceted technologies,
(3) Exchange ESR and ER among the different laboratories for short-term or longer-term, to promote their scientific and technical skills and to allow them to participate in postgraduate scientific and technology courses,
(4) Organize regular meetings with network participants and invited speakers in which ER and ESR can communicate with and present t heir work to an international forum of experts in the field and
(5) Train and promote transfer of knowledge at the interface of academia, industry and society.
