Evolution of the Protein-Interaction Networks: the SH3 network in Yeast

Objective

The overall goal of this Research Training Network (RTN) is to gain a quantitative understanding of the nature and evolution of protein interaction networks in eukaryotes. This will require the application of a broad range of methods and technologies, thus providing a unique opportunity for interdisciplinary training of early-stage and experienced researchers.

The RTN focuses on the study of the interplay between Src Homology 3 (SH3) domains, their host proteins and their binding partners (or target ligands) in four different yeast species, i.e. Saccharomyces cerevisiae (the best studied eukaryotic cell which exclusively profilerates by budding), Candida albicans (a human pathogen able to switch between bud-like and hyphal growth), Ashbya gossypii (a plant p athogen which exclusively grows like filamentous fungus), and Schizosaccharomyces pombe (the second best studied yeast which profilerates by bipolar growth and fission). These yeasts encompass a thousand million years of evolution.

The RTN intends to characterize the presently known interactions and to identify hitherto undetected interactions. Comparative analyses across the four genomes will allow us to investigate how new interactions were added to a network to cope with new necessities, as well as how t hey were lost when they were not needed anymore. The RTN will train researchers to be able to use a combination of approaches ranging from structural biology, biochemistry, genetics and genomics to advanced light microscopy and simulation tools for cell networks. To achieve these objectives a multidisciplinary approach will be carried out, by bringing together the expertise of leading research groups at a multilateral level.
