Combating cancer through novel approaches to protein-protein interaction inhibitor libraries

Objective

The objective of CAPPELLA is to develop a series of innovative small-ligand tools and libraries that allow new approaches to the inhibition of protein:protein interactions (PPI) in cancer.The inhibition of PPI is one of the most promising approaches to the development of novel cancer therapies. Because many PPI occur within the cell they can only be targeted by small molecules. But because PPI differ structurally from more classic drug targets such as enzymes and receptors, existing compounds have generally delivered disappointing results. New approaches are needed. The partners are "state of the art" in the research areas addressed. The project is a unique opportuniy to integrate novel insilico, chemical, genetic and ADME-based approaches to the design, synthesis and optimization of libraries and compounds. A key theme is the utilization of structural motifs found in natural PPI inhibitor compounds. This is coupled with high tent testing of the resultant structures on 3 distinct PPI targets relevant to different types of cancer, to allow compound rule-sets to be developed and improved. This project brings together some of Europe's leading biotech SMEs and several highly recognised academic institutes. By combining 5 distinct chemical approaches and testing them on 3 different targets (all from different partners) in three successive but repeating cycles the project mobilises resources from across Europe. All 9 partners have experienced teams and the coordinator's staff has experience of 7 prior EU projects. Each year there are about 10.9 million new cases of cancer and 6.7 million deaths. In Europe, 46% of cases are in women. By providing new approaches to a promising but challenging target class, perhaps 50% of cancer patients could one day benefit from the findings. The project provides the individual partners, and the European biotech industry, with tools and compounds having a high market potential and strengthening their competitive advantage.
