Objective
The threat of Bluetongue disease (BT) to European agriculture has stimulated the development of new vaccines against the virus. Existing vaccines to BT are based on attenuated strains of the causative virus. These vaccines are protective but cause viraemias, and teratological effects when pregnant animals are vaccinated. Consequently there is no way to distinguish between vaccinated and infected animals and the vaccines are not safe for use in all livestock. The aims of this project are to generate completely safe genetically inert vaccines for BTV, initially for serotypes that are identified in Europe with the future aim to generate a portfolio of vaccines that will protect against all BTV serotypes. To this end Merial has recently developed an inactivated virus-based BT vaccine effective against serotype 2 of the virus that has been already used in Europe although not in all potential ruminant hosts. Currently there is no data on the safety and efficacy of these vaccines in pregnant animals which will be realised in this proposal. As an extension of inactivated vaccine, LSHTM have developed a type of subunit candidate vaccine based on virus-like particles (VLPs) for US and Australian serotypes and validated these vaccines by demonstrating their protective efficacy in sheep in South Africa and Australia (see Roy's publications). On a previous EU project LSHTM recently constructed VLPs (that do not require passage of infectious virus during production) for European serotypes which are not yet validated in European breeds of sheep. Validation of these VLP will be performed, to demonstrate protective efficacy in sheep, cattle and goats. Tests capable of distinguishing vaccinated and infected animals will also be developed. In addition, the degree of cross-neutralisation achieved from vaccines containing mixtures of serotypes will be resolved. This proposal is targeted specifically at resolving each of these issues utilising complementary expertise within Europe.
