Analysis of heart development using gene trap transposon in zebrafish

Objective

We will use two lines obtained in a pilot gene trap screen to study heart development and function. The first line expresses a fluorescent reporter in neural crest-derived tissues, including outflow tract and atrio-ventricular valves and will be used to study the contribution of neural crest to heart development.

The second line expresses the reporter in the myocardium and has a transposon insertion into cardiac troponin T2 gene. It will be used to establish a zebrafish model of cardiomyopathy. We will also conduct an insertional mutagenesis screen in zebrafish using a novel, Tol2 transposon-based gene trap, with an aim to obtain 100 tissue-specific gene trap lines.
