Genomic approach to study the regulation and manipulation of energy-converting processes in model photosynthetic organisms

Objective

Life on earth depends on energy-converting processes between different biological sources. The primary and most important of these mechanisms is the conversion of solar into chemical energy that occurs in the vegetal kingdom. The aim of this project is to study basic regulatory signals in the energy-converting mechanisms of photosynthetic organisms employing state-of-the-art genomic and proteomictechniques having in mind its possible applicability. My plan of research is to study the regulation of proteins involved in photosynthetic and respiratory redo mechanisms that ultimately convert solar energy into high-energy phosphate bonds. A preliminary survey of the model plant species Arabidopsisthaliana has shown the presence of several is forms of pyrophosphates, glyceraldehydes-3-phosphatedehydrogenases, polyphosphates and other pip-dependent enzymes such as UDP-glucosepyrophosphorylase. Our plan is to study the expression of this genes at the RNA level, identifying expression patterns, and possible regulation by light, stress or circadian rhythms. At the same time, survey on T-DNA mutant collections will provide mutants in some of the genes that codify these enzymes and they will be tested for any phenotype associated to the above-referred patterns. Application of new high throughput genomic cloning techniques will allow the over expression of the proteins, fusion to cellular markers (GFP, LUC, GUS) and other tags to investigate their expression and regulation. Promoter analysis and yeast-one-hybrid experiments will also help to identify key-regulators of the processes which can then be manipulated to obtain better resource specimens. Our familiarity with another economically important model species, rice, will allow the application of these results to this regionally important crop and plans to contact local enterprises are due to happen.
