Objectif
The aims of the research is to determine biological techniques for the surveillance of workers exposed to emissions from coke ovens in order to assess the individual risk associated with exposure to a number of toxic and genotoxic substances. These biological surveillance techniques must be seen as an essential complement to measurement of the concentrations of hazardous compounds at workplaces. Furthermore, they must offer advances in the following areas:
-individual and collective means of protection, designed specifically to cope with emissions from coke ovens,
-the definition of medical criteria for the purpose of job applications or the retention of workers, in view of the multiple factors contributing to diseases suffered by coking plant workers.
The first phase of the research will be devoted to identification of the risk groups in the coking plants and the control groups (inside and outside the factories).
In the second phase of the research, the risk group in each of the coking plants involved will be the subject of a long-term systematic follow-up which will include on-site sampling (of atmosphere and of workers) and laboratory analysis.
The final phase of the research, which will be devoted to statistical analysis of the data obtained from the study of the risk groups and control groups, will make it possible to evaluate the reliability of these particular biological surveillance techniques.
METHODS AND MEANS BY WHICH THE AIMS ARE TO BE ACHIEVED
For the identification of groups a detailed questionnaire will be used which will take account of the factors affecting the genotoxic response: consumption of tobacco, consumption of medicines and of alcohol, sex ad age, nutritional parameters, type and quality of individual and collective means of protection.
The samples of gas and particles will be subjected to exhaustive quantitative analyses, covering organic and mineral aspects (gas chromatography chains - mass spectrometry, various techniques of atomic absorption spectrometry, etc.), in order to obtain a profile of the main elements believed to be genotoxic in the following chemical families: PAH, BTX, nitro-PAH, aromatic amines, heavy metals.
The biological study will include:
-an investigation of urinary mutagenicity with the aim of the AMES test;
-tests for chromosome aberrations on active lymphocytes,
-tests for sister-chromatid exchange on active lymphocytes,
-tests for mutagenicity of the gas and particle samples on human lymphocyte cultures.
