Objective
The aim of the research is to define the rome of infection in the development of lung disease associated with exposure to coalmine dust. The specific aims of the study are :
a) to determine the leukocyte response
b) to determine the effect if the immune response on a)
c) to examine the effects of low-level inflammation and endotoxin on a)
d) to determine the cellular response in pulmonary leukocytes in which an infection has been simulated.
Coalmine dust collected from the air of British collieries will be deposited in the lungs of rats under anaesthesia at the level at the bifurcation of the trachea.
Pulmonary inflammation will be assessed following a lapse of the required period of time by excising and lavage of the lungs to yield the leukocytes of the bronchalveolar space.
The immune response will be produced using a model antigen and the inflammatory response will be caused by a bacterial preparation.
The leukocyte response will be assessed in vitro in cells from treated and control rats.
METHODS AND MEANS BY WHICH THE AIMS ARE TO BE ACHIEVED
The leukocytes will be quantified by staining and differential counting.
To produce the immune response, the rats will be treated with a model of antigen (KLH) in Complete Freunds Adjuvant.
Inflammation will be produced by a heat-killed bacterial preparation of Corynebacterium paryum.
The following parameters will be measured to examine the leukocyte response : protease production and interleukin in I production
