The characterization of 3D flows is essential in developing novel flow devices involving process-intensification, separation, emulsification, etc. Flow characterization is commonly performed using particle image velocimetry (PIV), which is in general a 2D technique. Because many emerging flow configurations also have vertical velocity components (as e.g. in vortices or near flow distributors), the 2D nature of PIV is problematic for proper characterization. In this ERC-POC project, a recently introduced concept based on mechanical vibration of the sample across a confocal microscope was validated and implemented in a prototype instrument. The instrument was validated using an acoustic focusing setup that is used for separation of particles in a pharmaceutical context. A tracking algorithm was developed capable of properly tracking multiple particles simultaneously and the methodology was integrated in a user-friendly interface.
