Development and application of fine-mapping methods for the study of ETL in cattle

Summary: A bovine large-insert DNA library was constructed in a Bacterial Artificial Chromosome (BAC) vector. A total of 157,240 BACs have been picked into 384-well plates. Approximately 190 randomly selected clones were sized by Pulsed Field Gel electrophoresis, and have an average insert size of 105 kilobases. The library thus represents 5-6 genome equivalents and will give greater than 99% chance of recovering any given single-copy sequence. The frequency of clones without inserts is 4%.Clones from 360 x 384-well plates were gridded onto high-density nylon membranes. Superpools for PCR-screening were produced from the same set of clones.
