Objective
The principle objectives of this proposal concern the development of simple, inexpensive techniques to measure the level of cytokines and other functional proteins, and their specific mRNA, within defined cell populations using multi-parametric flow cytometry. The aim is to develop these tests into simple assays of clinical relevance. Research is required to develop and characterise the required fluorochrome conjugated anti-cytokine monoclonal antibodies and particularly to develop specific fluorchrome conjugated oligonucleotide probes to detect cytokine mRNA. It is also necessary to investigate and develop techniques for the culture and labelling of cells simultaneously with these reagents, to permit the concurrent flow cytometric detection of both protein and mRNA within individual cells. These assays have the potential to revolutionize the investigation of a wide range of diseases, both at diagnosis and during treatment.
