Objective
The low surface area-to-volume ratio offered by non-macroporous microbeads (microsupport with 3D geometry) to anchorage-dependent cells to grow into, makes it very difficult to reach cell concentration in excess of 5x10E6 cells/ml. The proposed Demonstration aims at validating at a cell concentration of 2,5x10E7 cells/ml NUNC's new generation of microsupport characterized by 2D geometry in 8L continuously stirred tank reactors. (Then also 20L). Supplementation with fresh nutrilites and simultaneous elimination of the toxics accumulated in these high cell density culture will be realized by sonoperfusion using the acoustic resonating wave technology developed by Anton PAAR GmbH. Validation of the latter acoustic separator (PAS) constitutes the other aim of the proposed Demonstration.
