Development of novel intelligent monitoring techniques and optimal adaptive control for fed-batch penicillin fermentation

Enzyme thermistors have a great potential for use as biosensors in fermentation and other process control systems. The thermistors are based on the measurement of the heat of enzymic reactions and can be used for offline and online analysis. They can be used to measure any product which reacts enzymically with a specific enzyme liberating heat which is converted in an appropriate electrical signal. A single system may be useful for monitoring various fermentations or other processes simultaneously for a given process parameter. Alternatively a set of in parallel systems can be used to monitor several different parameters. Interface electronics and software for improving the system performance and integration can be used. The advantages of the enzyme thermistor are its low cost and its specificity for the required analysis due to the enzymic reaction which is used.
Within the scope of the project a model has been developed which describes quite well the fed batch fermentation of penicillin. The structural properties of the model have been analysed and the yield coefficients identified. The practical applicability of the model developed on experimental pilot plant fermentations has been demonstrated in a great variety of conditions. The control strategy for the penicillin fermentation process which will be developed by the end of the project is expected to lead to increases in the yield of the product enabling a reduction in the production costs.
The SAMPLOR is an automatic sampling device which allows one to take automatically online samples from a fermenter or other reactor for further analysis. The samples are microfiltered through a ceramic membrane which is placed inside the fermenter and is steam sterilizable in situ. The ceramic membrane guarantees the sterility of the fermentation. This system has been largely experimented upon in low viscosity medium with very good results. It has also been used in viscous fermentation media with quite acceptable results. The stream from the sampling device can be fed to analysers which will perform the required analysis. The SAMPLOR is a very versatile apparatus which can be used for online sampling purposes on a great variety of fermentations or other reactions.
