Objective
IN THE PROPOSED EXPERIMENTS WE WISH TO IRRADIATE MOUSE FETUS IN UTERO WITH VARIOUS DOSES OF C060 GAMMA RADIATION AND EXAMINE BOTH THE DOSE DEPENDENT Tumor INDUCTION AND THE ACTIVATION 0F ONCOGENES (ras FAMILY, myc) AND INACTIVATION 0F TUMOR SUPPRESSOR GENES (P53, RETINOBLASTOMA PROTEIN) IN THE DEVELOPED TUMORS. DETAILS :
1. MOUSE FETUS WILL BE IRRADIATED IN UTERO WITH DIFFERENT DOSES 0F C060 GAMMA IRRADIATION AT DIFFERENT DAYS OF GESTATION (TABLE 1). TO MEASURE THE TUMOR INDUCTION MICE WILL BE SACRIFICED WHEN THEY LOOK VISIBLE ILL 0R AT TWO YEARS OF AGE. THE Tumor INCIDENCE 0F THE IRRADIATED AND THE UNIRRADIATED CONTROL MICE WILL BE COMPARED.
2. THE EXPRESSION OF THE K-, H-, N-ras AND myc ONCOGENES, AS WELL AS THE p53 AND Rb TUMOR SUPPRESSOR GENES WILL BE EXAMINED BOTH IN THE RADIATION INDUCED AND SPONTANEOUSLY DEVELOPED TUMORS.
3. THE AMPLIFICATION AND REARRANGEMENT oF THE ABOVE MENTIONED ONCOGENES, TUMOR SUPPRESSOR GENES WILL BE INVESTIGATED, TOO. 4. WE DETERMINE WHETHER THE ACTIVATION oF THE ras ONCOGENE FAMILY (K-, H- AND N-ras) AND THE INACTIVATION/ONCOGENIC ACTIVATION OF THE p53 PROTEIN ARE THE RESULTS OF POINT MUTATIONS IN THE CODING REGION OF THESE GENES. THE POSITION 0F THE MUTATIONS WILL BE DETERMINED BOTH IN RADIATION INDUCED AND SPONTANEOUS TUMORS.
5. COMPARING THE MENTIONED GENETIC CHANGES IN TUMORS OF IRRADIATED AND UNIRRADIATED MICE, WE INVESTIGATE WHETHER IDENTICAL ALTERATIONS OCCUR DURING THE RADIATION INDUCED AND SPONTANEOUS CARCINOGENESIS. WE ALSO EXAMINE WHETHER IDENTICAL CHANGES DEVELOP IN DIFFERENT TUM0R TYPES.
