Objective
METIER (MODULAR ECOTOXICITY TESTS INCORPORATING ECOLOGICAL RELEVANCE) ASSESSING THE TOXICITY AND BIOAVAILABILITY OF POORLY WATER SOLUBLE COMPOUNDS IN AQUATIC ENVIRONMENT WILL BUILD ON THE RESULTS OBTAINED FROM AN EXISTING EC STEP CONTRACT (EV5V CT91 0009).
THE GENERAL AIM IS TO CONTINUE TO DEVELOP A SERIES OF ECOLOGICALLY RELEVANT TOXICITY TESTS ALLOWING THE FULL BREADTH OF RESPONSE OF AQUATIC ORGANISMS TO CHEMICAL CONTAMINANTS TO BE ASSESSED UNDER REALISTIC EXPOSURE SCENARIOS. CURRENTLY, THERE IS AN URGENT NEED WITHIN THE EC TO INCREASE UNDERSTANDING IN THIS AREA, PARTICULARLY IN RELATION TO THE DEVELOPMENT OF REGULATORY GUIDELINES FOR POORLY WATER SOLUBLE COMPOUNDS (PRE NORMATIVE RESEARCH), WITH POTENTIAL INCLUSION ON A FURTHER AMENDMENT OF DIRECTIVE 67/548/EEC AND ON DIRECTIVE 88/379/EEC. HERE, WE WILL FURTHER DEVELOP OUR STANDARD FLOW THROUGH EXPOSURE SYSTEM FOR THE TESTING OF THE TESTING OF DIFFICULT SUBSTANCES, WHICH MAY BE USED FOR NORMATIVE PURPOSES, AND A SERIES OF TOXICITY TESTS LINKED TO A MECHANISTIC PHYSIOLOGICAL MODEL OF CONTAMINANT EFFECTS.
WE WILL CONCENTRATE ON POORLY WATER SOLUBLE COMPOUNDS, WITH AN EMPHASIS ON OBTAINING FURTHER INFORMATION ON EC PRIORITY 1 LIST OF CHEMICALS. THE RESEARCH IS DIVIDED INTO FOUR MAJOR SUBCOMPONENTS: (A) TECHNICAL DEVELOPMENT OF THE MODULAR TESTS SYSTEM, IN TERMS OF BOTH TESTS ENVIRONMENT AND TESTS ORGANISMS; (B) TECHNICAL DEVELOPMENT OF THE INDIVIDUAL BASED PHYSIOLOGICAL MODEL (ALTHOUGH DEVELOPING A POPULATION LEVEL MODEL IS CURRENTLY OUTSIDE THE SCOPE OF THIS PHASE, WE WILL START TO INCORPORATE HIGHER ORDER PHENOMENA) (C) DEVELOPMENT OF ECOLOGICALLY RELEVANT RAPID TEST ENDPOINTS (BIOCHEMICAL AND PHYSIOLOGICAL); (D) VALIDATION EXPERlMENTS TO TEST MODEL PREDICTIONS CARRlED OUT IN TEST MICROCOSMS (SPECIFIC AIM BEING TO DEMONSTRATE THAT SHORT TERM ENDPOINT EFFECT CONCENTRATION, WHEN PLUGGED INTO THE COMPUTER MODEL, WILL GENERATE PREDICTIONS THAT MATCH EFFECT CONCENTRATlON MEASURED IN THE VALIDATION EXPERIMENTS).
OUR SPECIFIC TASKS WILL BE TO PARTICIPATE IN SUBCOMPONENTS A,C AND d, BY USING THE MORPHOQUANT ANALYSIS ON CHLORELLA CELLS " MORPHOQUANT ANALYSER WITH SCANNING MICROSCOPE" PROGRAMME "MICROSCAN 80", AND PERFORMING ATP ANALYSIS IN CHLORELLA CELLS AND AT DAPHNIA MAGNA BY BIOLUMINESCENCE METHOD LUMINOMETER 1250 LKB3. ANALYSIS OF C,H,N,O AND S AT CHLORELLA CELLS AND DAPHNIA MAGNA ELEMENTAL ANALYSER CHNS/O, TYPE EA 1103, CARLO ERBA FISONS INSTRUMENTS. WE WILL ALSO PERFORM ADENYLATE ENERGY CHARGE ANALYSIS.
