Molecular Epidemiology of Diesel Exhaust Exposures

Objective



1) Alterations of heme metabolism in leucocytes as biomarker of exposure to diesel exhaust (ESTONIA)
The aim of this proposed project is the investigation of correlations between the alterations in heme biosynthesis and interaction of heme with DNA in leucocytes of workers exposed to diesel exhaust and the investigation of correlation between these changes of heme metabolism and the degree of exposure. This research will assist in identifying biochemical markers specific for diesel exhaust exposures in man. 2) DNA repair in mammalian cells damaged to chemicals which occur in diesel exhaust emissions (SLOVAKIA)
The aim of the project is to define the level of DNA, the mechanism of action and of some pahs and nitro pahs which are frequent in diesel engine exhaust. Attention will also be concentrated on DNA repair and induction of gene mutations and morphological transformation in mammalian cells. Cytochrome P450 plays a crucial role in the metabolic activation of many xenobiotics, including PAHs. Their role in the metabolic activation of nitro-pahs will also be studied.
