Objective
THE AIM OF THIS PROJECT IS: 1 ) TO ANALYSE THE EXPRESSION OF GROWTH-TRANSFORMATION-ASSOCIATED LATENT EPSTEIN-BARR VIRUS (EBV) GENES IN EBV-CARRYING B CELL NON-HODGKIN'S LYMPHOMAS (NHL) OF THE CENTRAL NERVOUS SYSTEM (CNS) OF AIDS PATIENTS, EITHER BY IMMUNOBLOTTING (IF SUFFICIENT FROZEN TISSUE FROM BIOPSIES OF PRIMARY CNS NHL IS AVAILABLE), OR USING THE METHOD OF RT-PCR (IF THE SIZE OF BIOPSY IS SMALL).
2) TO TAKE ADVANTAGE OF THE EXPERIENCE OF THE TEAM IN CONVENTIONAL PCR METHODS AND HISTOLOGY, TO SET UP THE METHOD OF IN SITU PCR. THIS WOULD ALLOW DETECTION OF VIRAL mRNA MOLECULES ON THE SINGLE CELL LEVEL ON FORMALIN FIXED AND EMBEDDED MATERIAL, ALLOWING TO EXAMINE A MUCH WIDER SAMPLE OF CASES.
3) IN ORDER TO CHARACTERIZE THE MOLECULAR GENETIC CHANGES POTENTIALLY CONTRIBUTING TO THE DEVELOPMENT OF B CELL LYMPHOMAS IN THE CNS, TO LOOK FOR REARRANGEMENT OF THE C-MYC PROTOONCOGENE (BY STANDARD SOUTHERN BLOT ANALYSIS, USING DNAS ISOLATED FROM TUMOR BIOPSIES), WHICH IS CHARACTERISTIC FOR BURKITT LYMPHOMA CELLS, TO ESTABLISH THE RELATIONSHIP BETWEEN THE PRESENCE OR ABSENCE OF C-MYC REARRANGEMENT AND THE EBV GENE EXPRESSION PATTERN .
INCLUSION OF DOCTOR GYURIS AND HER TEAM IN THE CONCERTED ACTION "PATHOLOGY OF THE NERVOUS SYSTEM IN HIV INFECTION" WOULD BE OF GREAT INTEREST BOTH FOR THE CONCERTED ACTION (CA) AND THE MICROBIOLOGICAL RESEARCH GROUP OF THE NATIONAL INSTITUTE OF HYGIENE OF BUDAPEST.
1) THE STUDY PERFORMED BY THIS TEAM WILL REPRESENT A VALUABLE COMPLEMENT TO THE RESEARCH ALREADY PERFORMED WITHIN THE FRAME OF THE CA BY PR.J.MIKOL AND PR.F.MORINET ON THE ROLE OF EBV IN THE PATHOGENESIS OF CNS NHL. IT WILL ALSO MARKEDLY INCREASE THE EXPLOITATION OF THE MATERIAL COLLECTED IN THE DEPARTMENT OF NEUROPATHOLOGY OF THE NATIONAL INSTITUTE FOR NERVOUS AND MENTAL DISEASE OF BUDAPEST BY DR.C.MAJTENYI WHOSE PECO PROJECT TO JOIN OUR CA HAS BEEN ACCEPTED IN 1993. 2) ON THE OTHER HAND, TO BE INCLUDED IN THE CA WILL ALLOW THE TEAM OF DOCTOR GYURIS TO COLLECT MORE RAPIDLY A "CRITICAL MASS" OF RARE MATERIAL SUCH A FROZEN MATERIAL FROM BIOPSIES OF PRIMARY CNS LYMPHOMAS IN AIDS CASES.
