Objective
The ongoing EC funded project is to construct an integrated physical, genetic and transcript map of Xq2l.1-q21.3 a region of approximately 15-20 megabasepairs (Mb). This work includes building contigs (continuous overlapping sets) of cloned DNA in Yeast artificial chromosomes (YACS)and smaller insert cosmid clones. These clones Will be mapped bY fluorescent in situ hybridisation (FISH) to a series of translocation and deletion breakpoint chromosomes in the region. The cloned DNA Will also be used to develop new highly polymorphic microsatellite genetic markers and to isolate new genes using several techniques most notably exon amplification and CDNA selection. There are several disease genes located in this region that Will be analysed in detail for candidate genes.
The proposal is to expand the existing project to include the laboratory of Dr Zajac from Bratislava. The research project would also be expanded to include tW0 chromosome rearrangements in the adjacent Xql3 region involved in tW0 cancers (acute myeloid leukemia and plexus papilloma). This is an important addition since Dr Zajac's laboratory is primarily interested in cancer genetics and Will in the future be able to apply the genome analysis techniques outlined in the present proposal to research and diagnostic approaches to a variety of inherited and acquired cancers. Although they have expertise in general molecular biological techniques they have not had extensive experience in the genome analysis techniques described above. The proposal is for Dr zajac to spend 9 months in the ICRF laboratory in Oxford to learn and aPPIY the genome analysis techniques described above on the Xq2l.1-q21.3 region. ThiS Will require funding for his travel to and subsistence in Oxford as Well as consumable costs for the laboratory expenses in oxford. Upon returning to Bratislava after 9 months he Would implement these techniques and train two students. For the two students We are requesting 12 months' salarv and consumahl + They Would continue work initiated bY Dr Zaiac on the Xal3 cancer causing chromosome rearrangements as Well as contribute to the Xq2l.1-q21.3 genome project.
