Objective
ELABORATION AND COMPARATIVE EVALUATION OF TEST SYSTEMS FOR THE ASSESSMENT OF GENETIC EFFECTS OF ENVIRONMENTAL CHEMICALS AND OF THE PRODUCTS OF THEIR METABOLIC TRANSFORMATION.
THE PROJECT COVERS THE FOLLOWING INVESTIGATIONS:
SALMONELLA TYPHIMURIUM SYSTEMS:
EXPERIMENTS WILL BE CARRIED OUT ISOLATE AND IDENTIFY THE PLANT MEDIATED ACTIVE INTERMEDIATES RESPONSIBLE OF THE GENOTOXIC ACTIVITY OF PESTICIDES, AS WELL AS METABOLITES IN HUMAN URINE AND BODY FLUIDS OF MAMMALS. SUCH ISOLATES WILL BE TESTED BY USING THE AMES-TEST.
ASPERGILLUS NIDULANS SYSTEM:BY USING TEST PROCEDURES WIDELY EMPLOYED AND VALIDATED FOR THE INDUCTION OF MITOTIC NON-DISJUNCTION IN A. NIDULANS THE ACTIVITY OF CHEMICALS ACTING ON TARGETS DIFFERENT FROM DNA WILL BE ANALYSED.
ONGOING WORK ON TESTING PROMUTAGENS WITH THE SO CALLED "GROWTH-MEDIATED ASSAY" PROCEDURE WILL BE CONTINUED BY TESTING DIFFERENT CLASSES OF PROMUTAGENS UNDER OPTIMAL CONDITIONS FOR THE METABOLIC ACTIVATION.
FURTHERMORE, EXPERIMENTS WILL BE CARRIED OUT IN ORDER TO ISOLATE AND CHARACTERISE A. NIDULANS MUTANTS ALTERED IN THE CELL WALL PERMEABILITY WHICH WILL BE CONFRONTED WITH WILD-TYPE STRAINS AS REGARDS THEIR SENSITIVITY.
DNA REPAIR STUDIES IN HUMAN LYMPHOCYTES:
THERE IS EVIDENCE FOR A CONSIDERABLE VARIABILITY AMONG NORMAL, HEALTHY SUBJECTS WITH REGARD TO THE UV-INDUCED DNA REPAIR SYNTHESIS. ON THE BASIS OF SUCH EVIDENCE EXPERIMENTS WILL BE CARRIED OUT TO TEST IF IN LYMPHOCYTES OF HUMAN POPULATIONS EXPOSED TO AGENTS FOR WHICH GENETIC EFFECTS ARE KNOWN, THERE IS A DEFINED ALTERATION IN THE ABILITY TO REPAIR UV-INDUCED DNA DAMAGE.
SPLIT DOSES STUDIES:
THE SPLIT DOSE PROTOCOL OF A GENOTOXIC AGENT MIMICS THE ACTUAL EXPOSITION OF HUMANS BETTER THAN TREATMENTS WITH SINGLE HIGH DOSES. EXPERIMENTS WILL BE CARRIED OUT WITH IN VITRO CELL CULTURES (3T3) TO FOLLOW THE EFFECTS OF DIFFERENT TIME INTERVALS BETWEEN DOSES ON THE INDUCTION OF MUTATION, DNA-REPAIR AND TRANSFORMATION. AGENTS WITH DIFFERENT DNA-REPAIR KINETICS AND MECHANISM OF ACTION WILL BE USED.
