STUDY OF THE MUTAGENIC AND TRANSFORMING EFFECTS OF ENVIRONMENTAL MUTAGENS, INCLUDING EXTRACTS FROM DIESEL EXHAUSTS

Objective

1) ELABORATION AND COMPARATIVE EVALUATION OF TEST SYSTEMS FOR THE ASSESSMENT OF GENETIC EFFECTS OF ENVIRONMENTAL CHEMICALS AND OF THE PRODUCTS OF THEIR METABOLIC TRANSFORMATION.
2) ASSESSMENT OF THE MUTAGENIC POTENTIAL OF DIESEL EXHAUSTS.

THE CONTRACT COVERS TWO PROJECTS:

1) IMPROVEMENT OF THE METHODOLOGY AND VALIDATION OF TESTS FOR MUTAGENIC ACTIVITY AND TRANSFORMATION TESTS.

THE OBJECTIVES OF THE PROJECT ARE TO PERFORM COMPARATIVE STUDIES BETWEEN MUTAGENIC AND TRANSFORMING EFFECTS OF ENVIRONMENTAL CHEMICALS AND SIMULTANEOUSLY TO CONTRIBUTE TO THE VALIDATION OF THE CELL TRANSFORMATION SYSTEM.

FOR THE TRANSFORMATION EXPERIMENTS TWO CELL TYPES WILL BE USED: SYRIAN HAMSTER EMBRYO CELLS (SHEC) WHICH METABOLISE ALMOST ALL KNOWN CARCINOGENS AND THE C3H/10T/1/2 ESTABLISHED CELL LINE METABOLISING ONLY PAH. THE CRIRETTERIA FOR THE EVALUATION OF THE EFFECTS ARE THE FORMATION OF FOCI AND THE FORMATION OF ABNORMAL COLONIES.

FOR THE EVALUATION OFTHE MUTAGENIC THE V-79 ESTABLISHED CHINESE HAMSTER CELL LINE WILL BE USED; THE INDUCTION OF MUTANTS, 6-THIOGUANIN RESISTANTS AND THE SCE WILL BE THE CRITERIA, COMPLEMENTED BY THE IN VIVO MICRONUCLEUS TEST..

2) MUTAGENIC AND CARCINOGIC EFFECTS OF DIESEL EXHAUSTS.

IN THIS PROJECT THE IN VITRO AND IN VIVO MUTAGENIC AND CARCINOGENIC POTENTIAL OF DIESEL PARTICULATES AND EXTRACTS USING SERUM AND ACETONE AS VEHICLES WILL BE STUDIED.

THE MUTAGENIC EFFECT WILL BE STUDIED USING V-79 CELLS FOR SCES ANS TGR INDUCTION IN VITRO, AND THE MICRONUCLEUS TEST IN MICE OR/AND IN CHINESE HAMSTER.

THE CARCINOGENIC EFFECT WILL BE EVALUATED

- IN VITRO, USING THE SYRIAN HAMSTER EMBRYO CELLS AND C3H/10T1/2 CELLS

- IN VIVO, INOCULATING SUB-CUTANEOUSLY THE PARTICULATES WITH OR WITHOUT VEHICLE.

AT A LATER STAGE, THE CARCINOGENIC EFFECT OF SOME SPECIFIC COMPONENTS OF THE DIESEL EXHAUST WILL BE STUDIED.
