NEW METHODS FOR THE EVALUATION OF COMBINED EFFECTS OF ENVIRONMENTAL AGENTS ON PULMONARY MACROPHAGES AS TEST CELLS

Objective

TO ESTIMATE THE DEGREE OF TOXICITY OF SELECTED SUBSTANCE, AND COMBINATIONS THEREOF, BY CONTINUOUS OBSERVATION OF CELL METABOLISM.

MICROCALORIMETRY IS USED FOR CONTINUOUS OBSERVATION OF CHANGES IN THE METABOLIC RATE OF RABBIT PULMONARY MACROPHAGES EXPOSED TO VARIOUS COMBINATIONS OF METAL PARTICLES AND OTHER SELECTED AGENTS.

THE HEAT PRODUCTION FROM THE CELLS IS TAKEN AS A MEASURE OF THE CELL MEABOLIC LEVEL.

A COMBINATION OF SEM AND ENERGY DISPERSIVE X-RAY TECHNIQUES IS USED TO DETECT THE METAL PARTICLES CONTENT OF INDIVIDUAL CELLS, AND THE NUMBER OF PARTICLES SMALLER THAN 2 OM.

CELL VITALITY IS ESTIMATED BY DETERMINATION OF THE LACTATE DEHYDROGENAS (LD) RELEASE FROM THE CELLS.
