Objective
TO DETERMINE USEFULNESS OF MEASURING MUTAGENIC AND CYTOTOXIC OF AIR POLLUTANTS BY VARIOUS TECHNIQUES.
THERE ARE FOUR MAIN PHASES:
1) 24-HOUR AEROSOL SAMPLES ARE TAKEN, USING HIGH-VOLUME-SAMPLERS. EXTRACTS ARE TESTED FOR MUTAGENICITY IN THE SALMONELLA STRAINS TA 98, TA 100, TA 89 NR AND TA 98 DNP. CYTOTOXICITY IN V79 CHINESE HAMSTERS ARE ALSO MEASURED.
2) SAMPLES OF AIRBORNE PARTICULATE MATTER ARE ALSO EXTRACTED AND TESTED AS IN 1) ABOVE.
3) CRUDE AEROSOL EXTRACTS ARE TESTED IN MUTAGENICITY ASSAYS OTHER THAN THE AMES' ASSAY, E.G. GENE MUTATIONS IN MAMMALIAN CELLS IN VITRO; GENE CONVERSION IN YEAST; DNA-REPAIR SYNTHESIS IN PRIMARY RAT LIVER CELLS; SEX-LINKED RECESSIVE LETHAL AND SOMATIC MUTATION TESTS IN DROSOPHILA.
4) DROSOPHILA MELANOGASTER FLIES OR LARVAE ARE EXPOSED IN ACTUAL POLLUTED SITES. EFFECTS ARE MEASURED BY THE SEX-LINKED RECESSIVE LETHAL TEST AND/OR THE WING-HAIR SOMATIC MUTATION TEST.
