ASSESSMENT OF HEALTH RISK FROM PRENATAL RISK FROM PRENATAL EXPOSURE TO GENETOXIC CHEMICALS

Objective

ELABORATION AND COMPARATIVE EVALUATION OF TEST SYSTEMS FOR THE ASSESSMENT OF GENETIC EFFECTS OF ENVIRONMENTAL CHEMICALS AND OF THE PRODUCTS OF THEIR METABOLIC TRANSFORMATION.

THE PROJECT AIMS AT ASSESSING POTENTIAL RISKS TO HUMAN HEALTH FROM PRENATAL EXPOSURE TO GENOTOXIC ENVIRONMENTAL CHEMICALS BY SHORT-TERM TESTS (MICRONUCLEUS, SISTER CHROMATID EXCHANGE, GENE MUTATION). THESE WILL BE APPLIED PRECISELY DEFINED CELL TYPES AND TAKE ACCOUNT OF THE PARTICULAR CELL-AND ORGAN-SPECIFIC EFFECTS OBSERVED FROM LONG TERN TRANSPLACENTRAL CARCINIGENESIS ANIMAL STUDIES AND INDICATED BY EPIDEMIOLOGY OF HUMAN CHILDHOOD CANCER. PARTICULAR ATTENTION WILL BE GIVEN TO BLOOD, AND LYMPHOID CELLS, AND THE DEVELOPING BRAIN. THE ACTIVITIES OF POTENTIALLY GENOTOXIC ENVIRONMENTAL CHEMICALS IN THESE IN VIVO/IN VITRO ASSAYS WILL BE COMPARED WITH THEIR MUTAGENIC POTENTIAL MEASURED BY SPECIFIC LOCUS IN VITRO ASSAYS BASED ON MOUSE LYMPHOMA L518Y CELLS, AND HUMAN LYMPHOBLASTOID CELLS, COUPLED TO CELL MEDIATED METABOLIC ACTIVATION SYSTEMS, INCLUDING THOSE DERIVED FROM PRENATAL ORGANS. EXPERIMENTS IN ALL THESE SYSTEMS WILL BE DESIGNED TO ENABLE CORRELATION TO BE MADE BETWEEN "GENETIC" END POINTS AND MOLECULAR LESIONS IN DNA AND CHROMATIN.
