Objective
DEVELOPMENT AND VALIDATION OF METHODS TO IDENTIFY NON-GENOTOXIC CARCINOGENS.
THE MECHANISMS OF CELL TRANSFORMATION ARE STUDIED WITH A NUMBER OF CELL SYSTEMS USING MUTAGENIC AND NON-MUTAGENIC CHEMICALS. THE BIOLOGICAL, ENZYMATIC AND GENETIC END-POINTS ARE COMPARED.
IN PARTICULAR, THE FOLLOWING TYPES OF STUDIES ARE CARRIED OUT:
I) PREDICTABILITY OF MUTAGENICITY-CARCINOGENICITY AND SHORT TERM SKIN TEST: MUTAGENS ALREADY STUDIED IN THE PROGRAMME AND A NUMBER OF RECOGNIZED PROMOTERS ARE TESTED IN VARIOUS MUTAGENICITY AND SHORT TERM SKIN ASSAYS. THEIR CARCINOGENIC POTENTIAL IS PREDICTED AND VERIFIED IN A LONG TERM SKIN TEST.
II) STUDY OF THE RESPONSE TO MUTAGENS/CARCINOGENS OF SENSITIVE AND RESISTANT STRAINS OF MICE: THE WORK INCLUDES CYTOGENETIC ANALYSIS IN BLOOD LYMPH AND EPIDERMAL CELLS.
III) VALIDATION AND DEVELOPMENT OF IN-VITRO CELL TRANSFORMATION SYSTEMS :
VALIDATION OF BALB/3T3 MOUSE CELL LINE AND SYRIAN HAMSTER EMBRYO CELLS. THE POTENTIAL OF THESE CELL SYSTEMS TO DETECT VARIOUS CLASSES OF CARCINOGENS WILL BE IMPROVED.
