Objective
This program proposes to study the freezability of various maturation stage bovine oocytes in view to define a suitable protocol for oocyte freezing. The interest of freezing oocytes is to manufacture banks of frozen gametes to preserve the gene line of species threatened with extinction, or to aid in many areas of research in zootechnology.
This research will be performed to evaluate oocyte potential sites of alterations : protein synthesis patterns, phosphorylation patterns, cytoskeleton organisation, intercellular coupling between oocyte and its surrounding cells after either rapid freezing or vitrification at different stages of maturation.
The survival of frozen oocytes will be thus checked by their ability to complete maturation,to be fertilized and further develop to the blastocyst stage, but also by their competence to manifest synchronous protein synthesis pattern changes or synchronous cytoskeleton reorganization as unfrozen control oocytes. This will be a guarantee of inocuousness for such a freezing procedure.
