Objective
Lower eukaryotes possess a multiplicity of transporter proteins (permeases) which exhibit different properties with respect to substrate specificity, affinity, velocity, capacity or regulation. In Aspergillus nidulans, three genes encoding proline permease (prnB), and two purine permeases (uapA and uapC) have been identified, isolated and studied in detail at the physiological, genetic and molecular level. The principal objective of this project is to use the Aspergillus nidulans transporter genes as an easily manipulated genetic, molecular and biochemical system to study the basic biological process of solute transport across the eukaryotic plasma membrane. A structure-function analysis will be made by selecting and constructing a large number of conditional, partial loss-of-function and specificity mutations, which will then be characterised at a molecular and biochemical level, in order to understand the role of different aminoacid residues in tranporter protein structure and/or function.
