In vivo and in vitro characterization of hptx3: A new member of the 'acute phase' pentraxin gene family.
ptx3 was recently cloned as an IL1-inducible gene in endothelial cells and belongs to the pentraxin gene family, including C-reactive Protein (CRP) and Serum Amyloid P Component (SAP). In spite of structural similarities (limited to the C-terminal half of the molecule) human ptx3 (hptx3) differs from classical pentraxins as for its genomic organisation, chromosomal localization and, more importantly, in vitro expression since it is not restricted to hepatocytes as for SAP and CRP and it is also evaluable in fibroblasts, monocytes and endothelial cells. In order to characterize the biological properties of the ptx3 protein, CHO cells have been transfected and a high producer recombinant clone has been selected. We plan to use the supernatant of this clone as a source of ptx3 to analyze the modulation of endothelial cell functions and the mitogenic activity of the protein. Furthermore, since ptx3 may represent the first identified member of a group of 'long' pentraxins (including the recently cloned apexin from guinea pig) we propose to identify new members of this 'long' pentraxin gene family.
