Objective
Cardiac actin is expressed during skeletal muscle development and in the adult heart. Transcriptional regulation of the gene depends on two enhancers located at -5 Kb and -7 Kb. The activity of the distal enhancer, in strengthening expression in the embryonic heart and skeletal muscle, depends on an E-box which binds myogenic factors and on an AT rich sequence that binds an Oct-l containing complex. Oct-l by itself does not bind to the sequence. This distal enhancer does not show MyoD transactivation in non-muscle cells, suggesting that MyoD family-independent and muscle-specific Oct-l co-factor(s) exist. The aim of the research project is to isolate and characterize the putative muscle Oct-l co-factor(s) by interaction cloning. The expression and the function of the co-factor will then be studied. The interest of the project is centered on the potential importance of an Oct-l containing complex in muscle-specific gene regulation.
