The role of connectivity in maintaining genetic diversity in nuphar lutea ; a model system for aquatic macrophytes in river habitats

Objective



The normalisation of large European rivers has resulted in fragmentation and loss of riverine habitats, resulting in a loss of genetic diversity within the remaining small populations through inbreeding and genetic drift, enhancing extinction risks. We will measure the genetic diversity as a function of population size and degree of isolation and establish the role of connectivity between populations by com- paring genetic patterns along and between rivers. This allows the construction of the phylogeography of a species which helps to support measures to maintain the genetic potential of aquatic macrophytes. The objectives of the exchange of a post-doc are. - To develop high resolution DNA markers for Nuphar lutea, our model macrophyte; - To standardize marking techniques between laboratories at Lille University (Fr) and at Nijmegen University (NL); - To measure genetic variation in 20 populations of N. lutea differing in degree of isolation, spread over two different river systems (Rhone and Rhine); - To construct the phylogeography of the sampled populations.
