Objective
TRANSCRIPTION ACTIVATION BY THE ESTROGEN RECEPTOR AND OTHER NUCLEAR RECEPTORS REQUIRES TRANSCRIPTION INTERMEDIARY FACTORS (TIFS). RECENTLY, A NOVEL NUCLEAR FACTOR (RIP140) HAS BEEN CLONED WHICH BINDS TO THE ESTROGEN RECEPTOR IN THE PRESENCE OF LIGAND. THIS INTERACTION REQUIRES A SHORT SEQUENCE IN THE LIGAND BINDING DOMAIN OF THE ESTROGEN RECEPTOR WHICH IS KNOWN TO BE IMPORTANT FOR ITS TRANSCRIPTIONAL ACTIVITY, AND WHICH IS CONSERVED IN ALL TRANSCRIPTIONALLY ACTIVE NUCLEAR RECEPTORS. THE OBJECTIVE OF THIS PROJECT IS TO ATTEMPT TO ELUCIDATE THE ROLE OF RIP140 IN TRANSCRIPTION, USING TWO APPROACHES. THE EFFECT OF RIP140 ON ESTROGEN RECEPTOR-MEDIATED TRANSCRIPTION WILL BE EXAMINED IN A RECONSTITUTED SYSTEM IN BUDDING YEAST, WHICH IS DEVOID OF ENDOGENOUS NUCLEAR RECEPTORS. THE YEAST TWO HYBRID SYSTEM WILL BE USED TO IDENTIFY ADDITIONAL PROTEINS ASSOCIATED WITH RIP140, AS BIOCHEMICAL EVIDENCE SUGGESTS THAT IT EXISTS AS PART OF A HIGH MOLECULAR WEIGHT COMPLEX WHICH MAY BE IMPORTANT FOR ACTIVATED TRANSCRIPTION.
